EGF概述
 

表皮生长因子(EGF)是由53个氨基酸组成的多肽,以1217个氨基酸残基的前体形式合成蛋白,经水解后发挥生物学活性,存在于唾液腺和人尿液提取物中[1]。EGF可诱导小鼠牙齿和眼睑的提前发育、抑制胃酸分泌、促进表皮的生长及角化过程。人EGF(hEGF)与小鼠EGF(mEGF)的多肽序列虽然在16个位点处不同,但两者的生物学活性基本相同,并能结合于同一膜受体。EGF及其受体信号也与肿瘤的发生和预后密切相关,EGF 受体抑制剂可用于小细胞肺癌、胃癌等恶性肿瘤疾病的治疗[1-3]

EGF的生物学功能
 

EGF通过与细胞膜上受体结合后激活多种酶发挥生物学作用,对炎性细胞、成纤维细胞表皮细胞及血管内皮细胞具有趋化作用,能够促进DNA、RNA 和羟脯氨酸的合成,调节蛋白质合成,完善胶原蛋白的构建,加快创面愈合速度[4]。研究发现EGF是一种强力的细胞分裂促进因子,有广泛的促进上皮细胞分裂增殖的作用,其以自分泌和旁分泌的方式作用于创面,促进表皮细胞及成纤维细胞等修复细胞的分裂、增殖,促进创面再上皮化,对血管生成也具有一定的诱导作用[5]。同时EGF还能刺激上皮细胞合成分泌胶原、透明质酸等细胞外基质,促进结缔组织细胞的生长,促进新生肉芽组织形成和伤口的再上皮化,加速皮肤、角膜及胃肠道等多种组织的创伤愈合,与肝的再生和创伤修复关系密切[6]

Bioactivity

Recombinant Human EGF (Cat. No. Y00801) stimulates proliferation of NIH3T3 cells. The ED50 for this effect ≤0.3 ng/mL. The acivity of Y00801 was equivalent to other competing products.
